Bone marrow adipogenic lineage precursors promote osteoclastogenesis in bone remodeling and pathologic bone loss

چکیده
Bone is maintained by coupled activities of bone-forming osteoblasts/osteocytes and bone-resorbing osteoclasts. Alterations in this relationship can lead to pathologic bone loss such as osteoporosis. It well known that osteogenic cells support osteoclastogenesis via production RANKL. Interestingly, our recently identified marrow mesenchymal cell population—marrow adipogenic lineage precursors (MALPs) form a multidimensional network bone—was computationally demonstrated be the most interactive with monocyte-macrophage through high specific expression several osteoclast regulatory factors, including Using an adipocyte-specific Adipoq-Cre label MALPs, we mice RANKL deficiency MALPs have drastic increase trabecular mass long bones vertebrae starting from 1 month age, while their cortical appears normal. This phenotype was accompanied diminished number attenuated formation at surface. Reduced signaling calvarial abolished osteolytic lesions after LPS injections. Furthermore, ovariectomized mice, elevated resorption partially MALPs. In summary, studies critical player controlling remodeling during normal metabolism pathological RANKL-dependent fashion.

The Role of Bone Marrow Aspiration and Bone Marrow Biopsy in Diagnosis of Bone Marrow Metastases
Background: Bone marrow is the site of many malignant disorders and it is one of the common places for solid tumors to metastasize. Examination of the bone marrow aspirate and biopsy is a routine procedure performed for assessment of various conditions such as cytopenias, hematologic neoplasms, nonmalignant disorders and metastatic neoplasms. Methods: The patients were referred to the Hema...
متن کاملThe farnesoid X receptor negatively regulates osteoclastogenesis in bone remodeling and pathological bone loss
Farnesoid X receptor (FXR, NR1H4) is a member of the nuclear receptor superfamily of ligand-activated transcription factors. Since the role of FXR in osteoclast differentiation remains ill-defined, we investigated the biological function of FXR on osteoclastogenesis, using FXR-deficient mice. We demonstrated that FXR deficiency increases osteoclast formation in vitro and in vivo. First, FXR def...

متن کاملBone cell precursors and the pathophysiology of bone loss.
In health, changes in bone formation and degradation rates are coupled and adequate cellular resources are available in the bone so that a change in bone formation rate occurs with an opposing change in resorption. On the other hand, the regulation of bone volume, particularly in pathological conditions, is dependent not only on the pathways that mediate terminal pathways of bone cell different...
متن کاملJaw bone marrow-derived osteoclast precursors internalize more bisphosphonate than long-bone marrow precursors.
Bisphosphonates (BPs) are widely used in the treatment of several bone diseases, such as osteoporosis and cancers that have metastasized to bone, by virtue of their ability to inhibit osteoclastic bone resorption. Previously, it was shown that osteoclasts present at different bone sites have different characteristics. We hypothesized that BPs could have distinct effects on different populations...
